G2L4 Reverse Transcriptase in DNA Repair by MMEJ
Biochemical and single-molecule assays for G2L4-mediated DNA repair; manuscript accepted in Nature Communications
Status: First-author manuscript accepted in Nature Communications; bioRxiv preprint
This project combines biochemical assays and high-speed atomic force microscopy (HS-AFM) to investigate how G2L4 reverse transcriptase participates in DNA repair through microhomology-mediated end joining (MMEJ).
Key Contributions
- Designed DNA oligonucleotide substrates with microhomology regions for DNA repair assays
- Optimized biochemical and single-molecule workflows to characterize reverse transcriptase-DNA interactions, repair intermediates, and ligase activity
- Visualized G2L4-mediated MMEJ and T4 DNA ligase activity using HS-AFM
